Several Positions Available - Biomedical Research PhD students and Postdoctoral Fellow CHROMATIN AND EPIGENETIC DETERMINANTS OF BIOLOGICAL AGE. • Competitive salary & comprehensive benefits package • Harry Perkins Institute & University of Western Australia The Terskikh laboratory discovered that patterns of epigenetic marks in the nucleus inform cellular identities and developed a novel technique: microscopic imaging of epigenetic landscapes (MIEL). Application of this technique to aging, termed image-based chromatin and epigenetic age (ImAge), captures intrinsic age-related progressions (trajectories) of the spatial organization of chromatin and epigenetic marks in single nuclei. ImAge represents the first-in-class imaging-based biomarker of aging with true single-cell resolution. The laboratory thrives on the crossroads of several interrelated fields and techniques, embracing a wide range of collaborations with leading researchers in the outstanding scientific environment of the Harry Perkins Institute, the University of Western Australia and worldwide (in particular, The Salk Research Institute and the University of California San Diego). The specific project(s) will be discussed with the candidate, but possible directions include: How does the aging process vary in different organs and cell types? To develop focused pipelines for quantitating biological age in different tissues at the single-cell level. To investigate and quantify the heterogeneity of the aging process in different organs and tissues at a single-cell level. To employ partial reprogramming in vivo using doxycycline-inducible expression of transgenic OSKM cassette to investigate the heterogeneity of in vivo rejuvenation at the single cell level. Testing the self-renewal vs disposable stem cell models of adult neurogenesis using lineage barcoding (e.g. PolyloxExpress reporter mice). To understand the implications for human adult neurogenesis for brain aging. The project can employ state-of-the-art confocal imaging, molecular and cellular biology approaches, including RNA-seq, ATAC-seq, Cut&Tag, Cut&See, and spatial OMICS at the single cell level, and state-of-the-art gene manipulations using RNAi and CRISPR technology. We routinely employ sophisticated mouse and human cell and tissue models. We utilize cutting-edge computational approaches, including machine/deep learning and generating custom-built algorithms and pipelines. Primary Responsibilities (depending on the project & PhD candidate vs Postdoctoral fellow) Conduct tissue culture and animal (mice) experiments as required by the project design. Use cellular and animal models to discover networks and pathways and to generate functional predictions and testable hypotheses. Independently design and perform experiments and procedures with a high degree of complexity, demonstrating increasing expertise with a wide range of techniques. Write reports, manuscripts, and grant applications with increasing responsibility over time. Knowledge, Skills, and Abilities (depending on the project & PhD candidate vs Postdoctoral fellow) Qualified candidates should be proficient in molecular and cellular biology, fluorescent microscopy, and gene expression/data analyses. Expertise in machine learning and familiarity with programming and code environments (Python, R, C, Jupiter notebooks) is desirable. Successful candidates should have demonstrated independent and critical thinking abilities and excellent communication and teamwork skills.
